Pharmacological Study of the Possible Effects of Temozolomide in the Treatment of Lung Cancer. Study of the Possible Effects of Temozolomide in the Treatment of Lung Cancer

Master Abstract

Lung cancer is one of the leading causes for death among other types of cancer and conventional chemotherapy pose extensive challenge including acquired cell resistance. The current study aimed to investigate the endotracheal inhalation (e.i) of temozolomide-loaded gold nanoparticles (TGNPs) or liposomes embedded temozolomide-loaded gold nanoparticles (LTGNPs) in treatment of urethane-induced lung cancer in BALB/C mice. Physicochemical characterization, zeta potential and electron microscope transmission were carried out for nanoparticle samples. In vivo study was performed for 19 weeks as follows (10 mice/group): group (1) untreated control; (2) treated with urethane (1.5mg/kg B.W, i.p) at day one and sixteen; group (3) treated with gold nanoparticles (GNPs100µ/kg B.w, e.i) once a week in the last three weeks; (4) treated with liposomes embedded gold nanoparticles (LGNPs 100µ/kg B.w, e.i) once a week in the last three weeks; groups (5) ,(6), (7) and (8) animals were pretreated with urethane at day one and day sixteen then treated with GNPs , LGNPs, TMZ-loaded gold nanoparticles (TGNPs,0.3mg/100µl/kg B.W, e.i) and liposomes embedded TMZ-loaded gold nanoparticles (LTGNPs,0.3mg/100µl/kg B.W, e.i), respectively once a week in the last three weeks. Lung and blood samples were collected for biochemical and histological examinations. The results showed that GNPs and LGNPs could be used as delivery system for chemotherapeutic agents. These findings revealed that endotracheal inhalation of TGNPs and LTGNPs are promising candidate for treatment of lung cancer due advanced localization of nanoparticles and drug targeting. Chemotherapeutic activity was pronounced in LTGNPs due to synergistic effect and increase time-release of chemotherapy
