A comparative biochemical study on the effect of polyphenolic antioxidants on NADPH oxidase and Nrf2 in Cisplatin-induced neurotoxicity in rats

Cisplatin (CIS) is an important chemotherapeutic agent used for the treatment of several cancers. However, various side effects such as neurotoxicity, nephrotoxicity, hepatotoxicity and ototoxicity restricted its usage for cancer treatment. Neurotoxicity is a clinically evident in patients that have undergone a full course of chemotherapy. This study is a trial to study the neuroprotective effects of some natural antioxidants curcumin and vitamin E as a powerful antioxidants against cisplatin–induced neurotoxicity in rats. This would be evaluated by estimation of some molecular parameters e.g, Nrf2, NQO-1, HO-1, GST1-α and NOX2 and some biochemical parameters such as enzymatic and non-enzymatic antioxidant e.g, catalase, glutathione, MDA as an evaluated markers in this treatment. The pretreatment with either curcumin or vitamin E caused a significant increase in the levels of Nrf2, HO-1, NQO-1, GST1-α and significantly decreased the higher levels of NOX2, NO and MDA when compared to cisplatin treated rats. Brain GSH level and catalase activity were significant increased in curcumin or vitamin E treated groups.
