Master Abstract

Abstract Chemical investigation of stems n-hexane fraction afforded ß-sitosterol and of the leaves ethyl acetate and n-butanol fractions afforded; kaempferol, quercetin, taxifolin, catechin, dehydrodicatechin A, gallic acid, quercetin-3-O-a-rhamnopyranoside, astragalin, quercetin-3-O-glucoside, Kaempferol-3-O-glucuronide, mequilianin, kaempferol-3-O-ß-D-(6''-n-butyl glucuronide), quercetin-3-O-ß-D-(6''-n-butyl glucuronide), kaempferol-3-O-ß-D-(6''-methyl glucuronide) and quercetin-3-O-ß-D-(6''-methyl glucuronide). The isolated compounds were characterized by their m.p., chromatographic behavior and different spectroscopic data (MS, UV, 1D and 2D NMR). Structural elucidation of these compounds was carried out by interpretation of their spectroscopic data and comparing it with the available literature. Quantitative Determination of Phenolics in different plant organs using HPLC analysis showed that: flavonols glycosides content were higher in all organs compared to the aglycones. Flowers and fruits are the most rich organs in while leaves, stems and bark are the richest in taxifolin and catechin. Total phenolics was estimated using Folin–Ciocalteu and total flavonoid using colorimetric assay. the results were in the order of the leaves = bark > stems > flowers > fruits with the values of 281.51, 280.60, 95.01, 74.71 and 27.88 µg GAE/g respectively for total phenolics and in the order of leaves > bark > flowers > stems > fruits with the values of 257.41, 217.21, 101.91, 87.45 and 53.93 µg RE/g, respectively for total flavonoids. Different fractions were also screened for their estrogenic and antimicrobial activity. Methylene chloride fraction was the most significant one for both estrogenic (increasing uterine weight) and antimicrobial (low minimum inhibitory concentration for all tested microorganisms) activity. Moreover, cytotoxic activity of the isolated compounds was evaluated against HePG-2 and MCF-7 cell lines using sulphorhodamine-B assay and doxorubicin as control. Quercetin showed the highest cytotoxic activity against the two cell lines (IC50 = 4.877 µg/ml and 0.8668 µg/ml respectively). Antioxidant activity in the different organs was estimated using DPPH assay and results showed that organs were in the order of bark > leaves > flowers > stems > fruits with the values of 450.30, 398.10, 369.91, 148.11and 2.95 µg AEAC/g, respectively.
